Cooke was educated at St Bede's College Manchester and Liverpool University. Cooke was honorary physician to Wigan Infirmary and medical officer of health for Aspull, near Wigan. Cooke was also in practice in Aspull with his father and brother, W.E. Cooke, and later his son E.P. Cooke. Richard Cooke died on 7 June 1954 aged 59.
