Mothersole was educated at Guy's Hospital, London. He spent his entire professional career in Bolton, where he was a consultant surgeon at Bolton Infirmary. He was a member of MMS and Manchester Surgical Society, and was chairman of the Bolton Division of BMA from 1922-6 and 1928-34, and was president of the Lancashire and Cheshire branch in 1932-3.
