Mellor was born in Coupland Street, Manchester, apprenticed to Fawdington, alongside Samuel Crompton (they were thought to look very alike). He was clerk to the physicians at MRI from 1830 to 1834. Between 1836 and 1838 Mellor gave anatomical demonstrations at Marsden Street Medical School in Manchester. He was a general practitioner in Burlington Street and later Chorlton-upon-Medlock. He was surgeon to the female penitentiary and later to Henshaw's Blind Asylum. He was president of MMS in 1868 and of the Lancashire and Cheshire Branch of the BMA in 1872. Mellor died in 1884 aged 76, at the time the oldest medical practitioner in Manchester.
