Mayne gained his medical education at St Bartholomew's before settling in general practice in Urmston, Manchester. Mayne was surgeon to the Manchester Warehousemen and Clerks' Association, and district surgeon to the Manchester Sheffield and Lincoln Railway Mutual Provident Association and Cheshire Lines County Railway.
