Knowles was born on 1 January 1884 and was educated at Leeds Girls' High School before qualifying in pharmacy. She spent some years as a dispensing pharmacist, during which time she developed her interest in medicine. She graduated in medicine at Leeds University and was house physician to the department of sick children at Leeds General Infirmary. Knowles then went into public health and became assistant medical officer in maternity and child welfare for Derby County Borough, Manchester and finally Leeds where she stayed until her retirement in 1948. Knowles died at Harrogate on 13 June 1975.
